چکیده

1. More than one mode of transport can be observed for concentrative uptake of a-aminoisobutyric acid (AIB) by rat kidney cortex slices when the initial concentration of AIB in the medium is varied between 0.2 and 16 mM. 2. The relative contribution from both systems to the total observed uptake at various concentrations of AIB was calculated. The K, values for the two systems were 3.87 rnM and 24.8 m&r. 3. Sodium-dependent inhibitory reactions influence the uptake of 0.8 rnM AIB more than of 8.0 mM AIB; inhibitors affecting -SH groups have the opposite effect. 4. L-Alanine is an effective competitive inhibitor of AIB transport at high substrate concentrations; more than one Ki value for L-alanine can be derived depending on the concentration of AIB. External L-proline enhances the affinity of AIB for uptake at low substrate concentrations; an inhibitory effect is observed at high substrate concentrations. Although omission of sodium from the medium abolishes concentrative uptake of AIB at all concentrations, it is restored at low substrate concentration by the addition of 4 mra L-proline to the external sodium-free medium. L-Proline placed internally stimulates AIB accumulation at low substrate concentrations only in the presence of sodium. 5. A site at which a ternary complex between proline or sodium and the AIB-carrier complex may be formed is proposed for AH3 transport at low concentrations; another system with different properties is used predominantly for AIB transport at high concentrations.

Discrimination of Single Transport Systems

Uptake of methionine, alpha-aminoisobutyric acid, and alpha-(methyl-amino)-isobutyric acid has been shown to occur by at least two transport systems, one sensitive and the other insensitive to the Na(+) concentration. For alpha-aminoisobutyric acid and its N-methyl derivative, the Na(+)-insensitive uptake is not concentrative and its rate increases almost linearly with concentration within the ...

Induction of amino acid transport in primary cultures of adult rat liver parenchymal cells by insulin.

Amino acid transport was studied in primary cultures of parenchymal cells isolated from adult rat liver by a collagenase perfusion technique and maintained as a monolayer in a serum-free culture medium. Amino acid transport was assayed by measuring the uptake of the nonmetabolizable amino acid, alpha-aminoisobutyric acid. Rat liver parenchymal cells transported alpha-aminoisobutyric acid by an ...

Methionine transport in S37 cells. Substrate-dependent function of amino acid transport system A in exchange processes.

Methionine had been observed to interact with two principal transport systems for amino acids in mammalian cells, the A and L systems. The present study of methionine transport and of exchange processes through system A arose in the course of a study to define the specificity of a transinhibition effect caused by cysteine. Methionine uptake through two transport systems in the S37 cell was conf...

Chloride dependent amino acid transport in the human small intestine.

Carriers of beta amino acids and imino acids in the small intestine of rabbits and guinea pigs are chloride dependent, and a cotransport of chloride, sodium, and 2-methyl-aminoisobutyric acid has been shown. This study examines the chloride dependence of amino acid transport in the human small intestine. The steady state tissue uptake of amino acids, given as the ratio between substrate concent...
